Malaria symptoms are caused by
toxins released into the blood as a result of the destruction of red blood cell
the multiplication of the malaria sporozites in the liver
the invasion of the red blood cells by the trophozoites
the development of merozoites into gametocytes

Once you're bitten by the female anopheles, the parasite enters the bloodstream and travels to the liver. The infection develops in the liver before re-entering the bloodstream and invading the red blood cells.
